ALKYLRESORCINOLS INFLUENCE ON HOMOSERINE LACTONES PERCEPTION IN THE LUMINESCENCE INDUCTION TEST OF THE ESCHERICHIA COLI LUXR + LUXI_LUXCDABE [№ 6 ' 2012] On model of a reporter luminescent strain E. coli JLD271, pAL103; luxR+ luxI_luxCDABE (A. Lindsay, B. M. Ahmer, 2005) we investigated influence of dormant state inductors — alkylresorcinols (AR) on bacterial perception of cell-density mediators — homoserine lactones (HSL). Reaction of bacteria to HSL didn't change by joint addition of AR, while use of preliminary processing mode by AR reduced the following luminescent induction by HSL in a concentration-dependent manner. Dependence of bioluminescence repression from AR molecules alkyl radical's length and hydrophobicity, also as suppression of a cellular metabolism are shown.

INVESTIGATON OF CARBON-BASED NANOMATERIALS BIOLOGICAL ACTIVITY IN BACTERICIDITY TEST ON ESCHERICHIA COLI AND BACILLUS SUBTILIS [№ 12 ' 2011] Influence of 10 carbon-based nanomaterials (CBNs): single- and multi-walled carbon nanotubes, nanofibres, С60-, С70-fullerenes, and amorphous carbon on viability of Escherichia coli K12 TG1 and Bacillus subtilis 168 is investigated. With use of named modeling microorganisms the CBNs bactericidal effect which progressed in time and depended on nanocarbon structural organization is shown. Comparison of test objects reaction shows B.subtilis little big sensitivity in comparison E.coli at high comparability of the biotesting results (r=0,927; P <0,005).

The results are discussed in the context of agarose films use as a test reference material to prepare an atomic force microscope for the further study of the mechanical properties of bacterial cells.

THE STUDY OF IONS, NANO- AND MICRO-SIZED CU AND FE PARTICLES BIOSAFETY IN TRITICUM AESTIVUM SEEDS GERMINATION TEST [№ 12 ' 2011] In Triticum aestivum seeds germination test the influence of equimolar concentrations of ions, nano- and micro-sized Cu and Fe particles has been investigated. The EC50 phytotoxicity level is much more exhibited on copper compared to the iron and dramatically increased at row line "microparticles > nanoparticles > ions". The most expressed reaction of wheat sprouts root system to influence of investigated toxicants is established.

BACILLUS SUBTILIS VKPM V-10548 STRAIN AND IT APPLICATION FOR SERUM BLOOD BACTIRICIDAL ACTIVITY RESEARCH [№ 12 ' 2011] The recombinant Bacillus subtilis strain carrying plasmid with gram-negative sea microorganism Photobacterium leiognathi luxAB genes, before each introduced gram-positive bacteria ribosome-binding site is designed. This strain characterized by high gene expression level of bacterial luciferase and was deposited in the Russian National Collection of Industrial Microorganisms under № V-10548. In researching of B.subtilis VKPM V-10548 reaction in contact with different component serum blood probes the correspondence between bioluminescence suppression and bactericidal effect level mainly determined by beta-lysine activity is shown.

СORRECTION OF RESULTS OF BIOLUMINESCENCT ANALYSIS IN VIEW OF OPTICAL PROPERTIES OF OBSERVED CARBONIC NANOMATERIALS [№ 6 ' 2010] A mathematical tool has been developed aiming at correction of results of bioluminescent analysis in view of optical properties of the examined samples. With its assistance a specialized computer application was created to quantitatively evaluate bioluminescence of bacteria existing in faintly clear (colored and/or blurred) environment. Use of this application while evaluating biotoxicity of carbonic nanomaterials allowed eliminating distorting effect of optical properties of observed compounds on the results of carried out analysis.
Romanenko N.A., Terekhova M.S., Deryabin ALKYLXYBENZOLS INFLUENCE ON FUNCTIONAL ACTIVITY OF FC-FRAGMENTS OF ANTIBODIES [№ 12 ' 2009] Ability of alkylxybenzols (AOB) change the functional activity of Fc-fragments of antibodies responsible for the initiation of the classical wais of complement activation is studied in the system of complementdependent hemolysis. It is determined that in such system C6-AOB and C12-AOB but not in C1-AOB and C3-AOB have their own haemolytic effect in high concentrations. At lower concentrations C6-AOB and C12-AOB influence on the kinetic characteristics of reaction of complementdependent hemolysis causing a time increase of lysis coming of 50% target cells without changing of lytic reaction speed.

INFLUENCE OF THROMBOCYTE CATION ALBUMEN ON BIOLUMINESCENCE AND VIABILITY OF RECOMBINANT STRAIN ESCHERICHIA COLI WITH CLONED LUX-OPERON PHOTOBACTERIUM LEIOGNATHI [№ 2 ' 2009] High sensibility of bioluminescence response of recombinant strain E.coli K 12 TG1 with cloned luxCDABE with gene P.leiognathi 54D10 to the influence of thrombocyte cation albumen (TCA) is established in this article. Determined for TCA quantities IC50, corresponding to 50-percent inhibition of luminescence intensity from starting level form in the same system 3,3-7,1 mkg/ml. It is shown that concentration dependences of bioluminescence suppression at the presence of TCA are inferior a lot to the development true bactericidal effect regarding to used bacterial cell-targets. As one of the possible reasons of such phenomenon it was named direct abscopal influence of TCA on enzymatic system of luminescence generation.

THE VARIANCE ANALYSIS OF THE ALKYL PHENYL ALCOHOL EFFECTS IN THE SYSTEMS "FERMENT-SUSTRATUM"AND "ANTIGEN- ANTIBODY" [№ 12 ' 2008] The significance of the alkyl phenyl alcohol (APA) impact on the reaction components "Ferment-Substratum" and "Antigen-Antibody" was quantitatively assessed through the variance analysis method. It was found that in these two-component systems the C1-APA acted both through modification of functional activities of the protein molecules and alterations of the substratum characteristics and, in particular, antigens. On the other hand, most significant action mechanism of the C6-APA was found to be its direct modifying impact on the ferment activity and antigen-binding capabilities of the antibodies.

PECULIARITIES RECOMBINANT LUMINESCENT BACTERIUM REACTION WITH DIFFERENT LUX-OPERONS IN PHAGOCYTAL SYSTEM [№ 12 ' 2007] Dependence of bioluminescent response of different recombinant strains Escherichia coli in phagocytal system from the nature of cloned lux-operons in them is determined in this article. It is shown that constructions’ using on the base of genetic material of sea luminescent bacterium Photobacterium leiognathi in interaction with neutrophils of peripheral human blood allows reaching direct proportionality between light intensity and evidence of killing effect developed in these conditions.

BASING OF OPTIMAL REGIME OF RECOMBINANT LUMINESCENT BACTERIA’S OPSONIZATION [№ 12 ' 2006] Optimal procedure of recombinant strain Escherichia coli K12 having luxCDABE gene P.leiognathi with using of normal human immunoglobulin in contrast to other potential opsonines (intact and thermoinactivated blood serum, complement) not giving bactericidal effect and minimal changed level of its initial bioluminescence concerning this microorganism is based in this article. It is shown that recombinant strains E.coli opsonizated with given way cause dose-dependent stimulation of luminol-dependent chemiluminescence of neutrophilic phagocytes but at the same contact themselves demonstrate progressive decrease of bioluminescence level the nature of which is determined admittedly with absorption and intracellular killing of bacterial cells.
